The present invention relates to a method capable of uniformizing a thickness of a sample which affects a pattern in a polymer MALDI spectrum and is capable of improving reproducibility of the MALDI spectrum, thereby enabling quantitative analysis of the polymer by MALDI mass spectrometry, and a method of manufacturing a sample for mass analysis of MALDI for quantitative analysis of polymer. According to the present invention, as the sample is applicable to a commercial MALDI-TOF equipment, quantitative analysis of the polymer can be performed more efficiently and quickly.
