Comparative Proteomic and Metabolomic Analyses to Study the Effects of Nanoparticle Exposure

Proteomics and metabolomics experiments have been conducted for plasma samples obtained from rats exposed to TiO2 NP's and from a control sample cohort. In proteomics studies, 12093 and 13253 separate peptide ion identifications correspond to 174 and 208 unique proteins obtained for samples from control and NP exposed animals, respectively. Comparative analyses indicate at least 29 proteins that are up regulated or down regulated upon exposure. Biopathway analyses using these differentially expressed proteins reveal a connection to systemic inflammation. The metabolomics analyses provided ~3800 dataset features observed to be significantly different between the two sample types. From these, exact mass matching and data filtering provides 29 metabolites that exhibit significant differences in abundance between the two sample types. Biopathway analyses of proteins linked to the identified metabolites reveal a connection to cell viability (cell death and survival).
