395 acetylated peptides from 144 mitochondrial proteins were quantified under three conditions by employing a quantitative proteomic strategy. A large percentage (31%) of quantified mitochondrial acetylated proteins illustrated metformin treatment-induced recovery effect. This highlights the importance of protein acetylation in various mitochondrial processes. This study not only extended the known inventory of in vivo acetylation sites and substrates in mitochondria, but also provided valuable information of the role of protein acetylation involved in the pathophysiology of type 2 diabetes as well as the molecular mechanism underlying the treatment of T2D using metformin.
