We successfully employed HDX-MS on virus-antibody assemblies in the molecular weight range of 3 to 9 million Da. As expected Fab binding reduces D incorporation at the known epitopes, the spike for Fab E1 binding & the baseplate for Fab 3120 binding. More interesting, regions distal to the epitope also undergo reduced D uptake, especially for the baseplate binding Fab (3120). This suggests that Fab binding induces a global rigidification of the capsid suppressing its breathing motions, even when only a few Fab are bound.
