摘要

Liquid chromatography/mass spectrometry is emerging because of its high specificity and multiplexing capacity. In this study, all "seven critical factors" [1] in LC-MS/MS protein quantification, including: internal standardization, protein purification, enzymatic digestion, proteotypic peptide selection, peptide purification, LC separation, and MS detection, have been optimized to obtain maximum sensitivity. Special focus was the selection of the optimal signature peptide, with respect to sensitivity and specificity.
