Histone acetylation is a dynamic modification that has been linked to numerous diseases, including neurodegeneration in brain tissue. Modifications of histones can affect interactions with DNA and, in turn, alter expression of genes. Changes in histone modifications are suggested to occur 20+ years prior to the development of symptoms of Alzheimer's Disease. Histone deacetylation is catalyzed by histone deacetylase (HDAC). 38 isoforms for HDAC1-11 with the majority of isoforms containing various deletions from the canonical sequence. Several histone deacetylases have been known to deacetylate non-histone proteins, such as HDAC6 catalyzed deacetylation of tubulin and HSP90 deacetylase. It may be possible that disease pathology includes non-histone targets by HDAC.
