摘要

A simple chemical deglycosylation procedure was developed to characterize o-glycan with three model glycoproteins. For each glycoprotein, the critical factor such as denaturing condition, alkaline concentration, and deglycosylation time need to be fine tuned to reduce degradation products and improve o-glycan release. Glcan profile obtained from TIC was comparable to that from FLD. Isomer forms of o-glycans were easily monitored by XIC and glycan composition can be verified by MSE fragmentation pattern.
