1. GLUT4/GLUT8 double KO (DKO) mice display region-specific alterations in the level of phospholipids in their brains compared with wild type (WT) mice. 2. These chan ges are the result of disru pted glucose homeostasis and alterations in lipid metabolism. 3. Both negative and positive modes were used to acquire comprehensive information on the alterations in phospholipids throughout the brain. 4. IMS confirmed that the hypothalamus, the nutrient sensing center of the brain, is one of t he distinctly different areas in the DKO mice. 5. IMS has provided additional information on the consequences of the DKO mutation on brain phospholipid and metabolite content. 6. Complete metabolic profiles will provide further insight into the roles of GLUT4 and GLUT8 in brain substrate metabolism and phospholipid content.
