Large Scale Analysis of RNA and Protein Expression Variation Through the Cell Cycle

Proliferating cells progress through deined cell cycle stages, which are characterised by events such as genome duplication and cell division. Each stage is associated with distinct biological functions, and transitions between each stage are tightly regulated to ensure high idelity replication. We have used a combined proteomics and transcriptomics approach to analyze the complex gene expression dynamics during interphase in a cultured human leukemia cell line (NB4). Whereas the vast majority of human cell cycle studies have relied on drug-induced synchronisation, we have used counterlow centrifugal elutriation, a physical method of separating cells of diferent cell cycle stages without drug treatment. Synchronisation by elutriation, which separates cells based on size, exploits the fact that cell size is cell cycle-dependent.
