Analysis of Testosterone and epitestosterone in male serum samples by way of 2D LC/MS/MS

The separation and quantitation of testosterone and epitestosterone from serum can provide an indicator for possible metabolic abnormalities. Traditional methods using either GC/MS or LC/MS require derivatization or fail to provide adequate separation of the epimers. The procedure described herein shows a rapid and sensitive method for the determination of testosterone and epitestosterone from serum samples utilizing a 2D LC/MS/MS approach. The resulting method affords full baseline separation of the epimers and excellent detection limits (less than 0.2 ng/mL in serum) within a short run time (7 minutes).
