1. The use of a reproducible and reliable analytical platform for biomarker discovery in neurological disorders using only 100 μL of CSF has been demonstrated. 2. The effect of pre-analytical variables was systematic characterized for the CSF proteome. 3. Time on bench after lumbar puncture causes higher number of changes in the composition of the proteome followed by temperature. Addition of protease inhibitors does not improve proteome integrity. 4. No proteomic differences were observed when lumbar puncture was performed with subjects sitting vs. recumbent and in a fasted vs. fed state with LPs conducted one month apart. 5. Significant proteome level differences were found in CSF due to aging among healthy individuals. 6. Much overlap in CSF protein changes were found between presymptomatic and symptomatic FAD. Results are consistent with inflammation and synaptic loss.
