The physical principles that determine ion mobility in a linear drift cell are well understood (1); consequently, collision cross sections can be directly calculated from the measured drift time having defined the experimental conditions. Such calculations are not possible for Travelling Wave (T-wave) mobility instruments due to the range of factors that influence ion motion through the mobility cell. Calibration of these instruments is thus required for each set of experimental conditions so that T-wave mobility experiments can be used to calculate analyte ion collision cross-sections. Here we describe the design of a universal mobility calibration standard, QCAL-IM, enabling empirical calculation of peptide collision cross-sections using a T-wave mobility instrument.
