The use of Ion Mapping scan functions w/ spatial LC-MS analysis was demonstrated to be a powerful analysis technique. A TriVersa NanoMate was used in LESA mode to successfully identify over 900 proteins from a single HPTLC development lane. An intact protein, myoglobin, was developed and readout using the Camag TLC-MS interface. Future Work: (1) Incorporate ion mobility into spatial LC-MS experiments and direct infusion experiments. (2) Develop higher quality HPTLC media that will better perform w/ MS techniques. (3) Address the ability to use ion mapping scan functions and the ability to obtain quantitative data. (4) Use multiple lane scans and MS experiments w/ the CF-LMJ-SSP to increase sequence coverages. (5) Study different peptide and protein development conditions on ProteoChrom HPTLC Silica gel 60 F(254S) plates.
