to decrease handling time, provide online analysis and perhaps increase resolution, a FPLC column will be custom packed and tested; this should allow for a uniform flow rate as well as an increased amount of theoretical plates, both of which favor a better separation. overall this protocol shows the potential to extract glycopeptides from protein digest samples with little peptide carryover; future work will concentrate on improving gradients and buffer compositions so as to enhance the extraction. moreover, neutral reporter molecules need to be tested on the derivatized and underivatized stationary phases, so as to determine the efficiency of derivatization. lastly, more complex samples need to tested, including those containing sulfate and phosphate moities so as to assess polar vs. WAX interactions.
