Proteomic Analysis of ePFT-Collected Pancreatic Fluid to Investigate Chronic Pancreatitis

We compared the proteomes of pancreatic fluid from chronic pancreatitis patients and controls. (1) We identified a total of 1392 different proteins in 18 pancreatic fluid samples. (2) Of these proteins, 257 and 413 were identified exclusively in the NP and CP cohorts, respectively. (3) Of the 722 proteins that were identified in both cohorts, spectral counting and statistical analysis thereof (QSPEC) revealed 41 and 93 proteins that were statistically up- or down-regulated, respectively, in the pancreatic fluid from individuals with chronic pancreatitis. (4) GoFact analysis revealed that most differentially-expressed proteins identified were extracellular (potentially secreted) and exhibited peptidase activity. (5) KEGG pathways analysis revealed that proteins in cellular metabolic pathways were fewer in number in CP verses NP, while those pathways linked to various diseases were more prevalent in the CP than the NP cohort. Our workflow resulted in the identification of potential biomarkers of chronic pancreatitis and provides a high-throughput approach for the further study of the pancreatic fluid proteome.
