Taking full advantage of dual ion funnel technology, it was possible to quantify underivatized estradiol at the low pg/mL level. Sensitivity of the method in phosphate buffered saline was demonstrated with LLOQ at 1 pg/mL underivatized E2 using a matrix-stripping LC approach capable of concentrating a 100 (mu)L injection volume to avoid band broadening effects. Necessity for chemical derivatization is eliminated utilizing the described system, alleviating the need for laborious preanalytical manipulation. This high quality estradiol assay with LLOQ velence 1 pg/mL and excellent performance will be an important tool for clinical research where assay sensitivity is one of the most important factors.
