The optimized sample preparation method used a one-step liquid-liquid extraction for sample preparation and consumed a small volume of sample (50(mu)L). The extraction method has a extraction recovery of 89.5percent-97.3percent and low matrix effect of less than 10percent. The LLOQ is 0.1 ng/mL and the linearity is excellent over the concentration 0.1-100 ng/mL. This validated assay demonstrated adequate sensitivity, accuracy, precision, and stability to support the analysis of rat plasma samples in pharmacokinetics study.
