MALDI-TOF-MS is used for TAG-estolide analysis, which offers a fast identification and characterization method with small sample volume consumption. MALDI matrices were optimized for TAG-estolide detection. The addition of sodium salt during matrix preparation significantly improved the signal intensity of TAG-estolides. DHB and THAP, with the addition of sodium salt, offered clean MS spectrum (MS mode), while CHCA offered the highest overall signal intensity and was selected for MS/MS spectra collection; LC-MALDI-MS was employed to separate and detect TAG-estolides from different oils, analytes with a 2Da mass difference can be separated and identified afterwards; The molecular mechanisms which result in the formation and degradation of TAG-estolides are currently under investigation.
