Several different methods have been applied in surface analysis of lipid molecular species such as MALDI, DESI, and LESA (liquid extraction surface analysis with NanoMate). Among several methods of surface analysis, MALDI is most popular and with significantly higher resolution and sensitivity. But concerning to MALDI there are some difficulties in ionization, such as detection with different adducts such as proton, sodium and potassium for single molecular species, and low ionization efficiency of minor components were often observed. In this experiment we tested LESA system combination with NanoMate as a new approach for the localization analysis of lipid molecular species in mouse brain tissue slices. Also we compared the experimental results with data obtained by the laser microdisection (LMD) combination with Nano-ESI. Addition to this we tried direct analysis of body fluid such as human saliva and mouse plasma on segmented membrane by LESA.
