The results confirmed both our expectations: (a) that normal BM should contain significantly more intact CXCL12 than BM from PV patients and (b) that while BM from normal subjects does not have contain truncates from CXCL12, all known truncates due to proteolyctic enzymes do appear in the BM of patients with PV, suggesting that a similar proteolytic environment exists both in the marrow and PB. The results support our hypothesis that the truncation forms of CXCL12 likely compete with the full length CXCL12 for CXCR4 expressed by CD34+ cells leading to the release of such cells from the marrow and their constitutive mobilization.
