Delipidation of ApoB48 and ApoB100 for Fast Quantitation by LC-ESI/MS/MS

SDC is an effective delipidation reagent for apolipoprotein B in plasma and required for complete digestion of the protein. 0.5percent SDC was determined to be optimal for these studies. This method allows absolute quantitation of apoB at concentrations as low as 30nM in mouse plasma, with method linearity demonstrated up to 10uM apoB in mouse plasma. At 9 minutes per LC/MS analysis, the method can be used to process one 96 well plate in 15 hours for a total of 1100 samples per week.
