Investigation of sub-cellular localization of molecules is important to clarify the cellular phenomena. On allergy, it is familiar enough that mast cells secrete chemical mediators by granule popping. That means molecules in granule are very important for allergy, however, exhaustive molecular analysis of granule contents has not performed yet. So we analyzed granules in RBL-2H3 cell line as a mast cell model which has the granules including chemical mediator such as histamine or serotonin. We succeeded to trap just one granule from a single cell by suction with nanospray tip under microscopic observation. Many specific ion peaks were detected in a granule. These molecules are under identification in relation to allergy mediators.
