This study demonstrated that thermal gradient focusing ESI can be used to develop microflow discovery PK assays with enhanced sensitivity without increasing chromatographic run times. This study also demonstrated that method migration to microflow conditions is relatively simple when columns with the same stationary phase and particle size are available if delay volumes are minimized. In addition to extending the LLOQ from 0.25 to 0.125 ng/mL, the microflow PK assay exhibited better accuracy and precision that the comparable narrow-bore method.
