Application of Dried Spot Sampling Technique in Cerebrospinal Fluid Sample Analysis

The assay achieved an LLOQ of 1.22 ng/mL. The assay was robust to allow for verification over 3 days of testing and met acceptable criteria for both intra- and inter-day precision. PK data using the CSF spotting correlated with historical data. DMPK-B card showed acceptable sensitivity, linearity, and selectivity for assessment of PHA-543613 in vivo. Future work will be evaluating other DMPK cards, DMPK-A and DMPK-C (untreated) and also verifying robustness with other compounds. In addition stability on card will be tested.
