EXTENDING LC-MS/MS LINEAR DYNAMIC RANGE FOR QUANTITATIVE ANALYSIS OF DRUGS IN DRIED BLOOD SPOT SAMPLES

A novel LC/MS/MS method was developed for antiretrovirals (Saquinavir, Indinavir and Ritonavir) in dried blood spots using QTRAP~(~R) 5500. Calibration curve ranging from 0.05 ng/mL to 10000 ng/mL, a 5.5 order magnitude was demonstrated accurately and precisely (acceptable limits +- 15percent) in dried blood spots using STDs and QCs. The new SignalFinder~(TM) Algorithm in MultiQuant~(TM) 2.0 extended the linear dynamic range from 4 orders to 5.5 orders. Dilution integrity in dried blood spot by extending LDR using MultiQuant~(TM) 2.0 software has been demonstrated using STDs and QCs. The need for further diluting high concentration samples could be replaced by detector saturation correction and demonstrated using QCs and has to be further explored using incurred samples.
