1. Successful in-house staple peptide synthesis and analysis by tandem mass spectrometry is demonstrated. 2. Radical stabilization and sterical effects can be used to rationalize the ECD PIA distribution of peptides containing alpha,alpha(velence)-disubstituted amino acids, similarily to ECD of beta-peptides. 3. The amphipathic nature of the BH3 peptide gives rise to a periodic ECD PIA in agreement with previous data. However that feature made it difficult to observe specific product ion yield modulation that would relate to the staple-induced structural change.
