Endogenous matrix components extracted from DBS cards alone causes significant matrix effect for both normal phase and reverse phase chromatography. It adds a great challenge to assay development, especially for hydrophilic compounds. The potential for samples stored in DBS format to have a much improved stability over samples stored as blood/plasma format is of interest to all bioanalytical scientists. Improved stability is especially beneficial to some compounds with challenging stability issues using conventional blood/plasma sample formats. The DBS sample drying process could potentially affect the recovery and assay performance and is worthy of further investigation.
