We have used SILAC and anti-pTyr peptide IP to profile in vivo changes in tyrosine phosphorylation sites in response to EphB receptor activation. This study not only provides a set of proteins as potential effectors in the EphB signaling pathway, but also more importantly reveals specific pTyr sites that may play important roles in regulation of the signaling. The link that is established between individual pTyr sites and the signaling will greatly facilitate subsequent functional investigation of the involved effector proteins.
