High Performance Liquid Chromatography Combined with Multi-stage Mass Spectrometry Analysis of DNA Adducts of Aristolochic Acids

A total of 4 DNA-AA adducts were completely separated and identified, including dA-AAI, dG-AAI, dA-AAII and dG-AAII. MS~(1) to MS~(5) spectra were acquired for all of the four adducts. The MS~(n) fragment ions obtained in LCMS-IT-TOF system shows different features from the previous results got by LC-TOF or LC-QTOF. The MS~(n) fragment ions were analyzed systematically; tentative structures of multi-stage fragment ions and fragmentation pathways were proposed. The MS~(2) spectra of the DNA-AA adducts displayed characteristic fragmentation pattern with loss of 116 Da, which was due to the neutral loss of a deoxyribose moiety. More structural information of DNA-AA adducts was obtained from the MS~(3) to MS~(5) spectra, which exhibited interesting features.
