We were able to observe distinct spectral differences in ion intensity, in number of peaks present, and between positive and negative MALDI ionization mode. Additionally, the very high mass accuracy obtained (approx100ppb at mass 700) provided the requisite specificity for the elemental composition determination. Our positive mode MALDI data from the use of THAP matrix, which is one of the common MALDI matrices utilized for lipid analysis, resulted in very good clustering of pre-and post- ERT GD patient samples. These small molecule profiling provides additional biomarker to our proteomics profiling of similar GD patient samples. The use of MALDI FT-ICR MS provides an excellent tool for fast data acquisition and profiling of complex small molecule mixture.
