A Label-free Approach to Quantitate Novel Ectopic and Intrauterine Pregnancy Serum Biomarkers

Label-free quantitative comparisons using the Rosetta Elucidator System for biomarker discovery and label-free validation with MRM are cost-effective alternatives to stable-isotope labeling-based quantitation methods. When entire proteomes were compared, several proteins were identified as higher, and a larger number of proteins were consistently lower in ectopic pregnancy compared with intrauterine pregnancy. GeLC-MS/MS provides the ability to identify and quantitate various isoforms of a protein which may be specific to ectopic pregnancy. Multiple ectopic pregnancy biomarkers have been discovered and validated on a small cohort; the next step is to expand validation to an independent set of samples.
