Invasion of the basement membrane is believed to be a critical step in the metastatic process. Ivlelanoma cells have been shown previously to bind distinct triple-helical regions within basement membrane (type IV) collagen [1]. Additionally, tumor cell binding sites within type IV collagen contain glycosylated 5-hydroxylysine (Hyl) residues. It has been previously reported that the type IV collagen al(IV)382-393 sequence interacts with the α2β1 integrin [2], and the al(IV)531-543 sequence binds to the α3β1 integrin [3]. In the present study, we have utilized triple-helical peptide models of the type IV collagen al(IV)382-393 and αl(IV)531-543 sequences to investigate the influence of glycosylation on adhesion and spreading of four different melanoma cell lines.
