Culture media optimization can lead to significant increases in product titers from recombinant CHO cell lines. When faced with limited development time for optimization, rigorous selection of methods and scales must be applied. A media optimization strategy has been developed to meet this need. Overlapping phases allow flexibility in optimization of process conditions, basal media, supplements, and feed strategies. A case study shows culture media optimization for two CHO cell lines using this process. Growth and productivity of both cell lines was significantly increased when compared with starting media. These results show that productivity can be rapidly increased using this methodology that hinges on the coordination of appropriate methodologies and scales during each phase of the optimization process.
