Nanoflow HPLC, coupled to an ion trap mass spectrometer, provides excellent peak shape and sensitivity for quantifying dA-AL-I adducts. The limit of detection was less than 50 fg of the adduct injected onto the column. Calibration curves were constructed over the range of 50 fg to 10 pg with excellent statistical correlation (R2 ≥ 0.99). Our initial experiments have detected the dA-AL-I adduct in kidneys from mice treated with AA-I. Data are presented showing adduct levels measured at selected doses and for varying time points after treatment.
