1. Targeted intact protein LC-MS/MS method development with recombinant protein substitutes provide initial understanding of biomarker behavior but may be influenced by presence of tags and other modifications. 2. Immunopurification of native PBP2a target protein from MRSA lysate facilitated accurate target precursor mass identification, selection, and fragmentation parameter selection. 3. The native PBP2a protein exhibits similar fragmentation characteristics as the recombinant protein and additional sequence tag ion series from the N-terminal region. Further optimization and alternative fragmentation approaches will make detection of this resistance marker protein feasible in more complex matrices.
