The work presented here describes the development of a polarity switching MSI method for untargeted screening of spatial distribution and relative abundance of metabolites in tissue sections. The developed method was used to investigate the differences in spatial distribution and relative abundance of more than 500 metabolites in healthy and cancerous hen ovarian tissue sections. Glutathione was chosen for quantitative analysis based on its increased abundance in cancerous tissue. A quantitative MSI method was developed to quantify endogenous glutathione in the cancerous tissue. A calibration curve consisting of SIL GSH was spotted directly on the tissue allowing direct correlation of SIL GSH abundance to native glutathione for absolute quantification in tissue.
