Bottom-Up Proteomic Analysis of Single HCT 116 Colon Carcinoma Multicellular Spheroids

The spheroids are accurate and precise biological replicates for mass spectrometry analysis, a property useful for high-throughput mass spectrometry experiments. The next step is to further investigate the inner workings of single spheroids by separating the layers and analyzing via mass spectrometry. This will be accomplished using serial trypsinization to gently dissociate the layers, followed by Filteraided sample preparation (FASP) for mass spectrometry analysis.
