Relative quantitation on thousands of identified protein groups demonstrated that significant changes caused by radiation exposure. As expected, higher radiation doses caused more severe damages as represented by number of affected proteins increase from 7 to 22% when 8Gy and 14Gy were compared. As radiation dose goes up, more proteins were down-regulated compared to the control indicating overall damages in protein activity and function. When up-regulated proteins were subject for the GO enrichment analysis, response to wounding was most activated cellular process whereas the down-regulated proteins were mostly involved in RNA processes. One protein of interest, Inter-α-trypsin inhibitor (ITIH4), which is known to be involved in connective tissue damage, was found to be increased by radiation induction and might play a role in promoting the fibrosis of lung.
