New methods for generating intact peptide radical cations using ion/ion interactions have been demonstrated using both collisional activation and UVPD. Generation of radical cations via UVPD has been carried out by homolytically cleaving C-I bonds on reagents that have been bound to peptides in the gas-phase both covalently and electrostatically. Collisional activation of azo functionalities has been used after ion/ion reactions were used to create both covalent and electrostatic products in the gas-phase. Activation of Direct Red 81 yields a stable radical, allowing for radical generation prior to interaction with the peptide.
