Staphylococcus aureus systemic infections cause abscesses whose size and severity worsen over the course of infection. IMS reveals proteins that are more abundant in abscessed regions or surrounding tissue. High mass resolving power instrumentation allows for the successful study of modified proteins by providing both mass accuracy and resolving power to successfully distinguish oxidized species. Protein identification reveals a cysteic acid modification of S100A8, a subunit of the neutrophil-associated protein calprotectin, that localized to infectious foci. Oxidative modifications to S100A8 could impact biological function and require further study to provide new insights into the pathogen-host interaction.
