Absolute quantitation of aminoglycoside antibiotics in mouse plasma by a HILIC-based LC-MS/MS method

Robust and reproducible high and low sensitivity LC-MS/MS methods were developed for quantitation of aminoglycosides in plasma to support screening PK studies in mice. Good chromatographic retention and separation of analytes were achieved by gradient elution using hydrophilic interaction chromatography (HILIC) on a Cogent Diamond Hydride column. SRM of doubly charged ions was applied to achieve high sensitivity; SRM of singly charged ions was used to de-optimize method sensitivity.
