Extensive characterization of photo-stressed AMC samples was performed with intact MDP analysis and LC/MS peptide mapping combined with a range of other assays. Mass spec analysis suggests increased modifications in AMC, especially Met and Trp oxidation in the MAb; some of these modifications may be responsible for observed decrease in binding affinity. The results derived from the extreme light exposure conditions facilitate building the foundation for choosing the appropriate analytical techniques for assessment of changes under normal storage conditions.
