摘要

MCO reactions were successfully used in the determination of metal-protein dissociation constants. Measured affinities were found to be comparable to previously-reported values. This MCO/MS method has utility in concurrently identifying metal binding residues and measuring affinity for the metal ion. Initial studies show promise for (a) evaluating complexes with very low dissociation constants and (b) systems with multiple metal binding sites.
