Enhancements in Analysis and Imaging of Small Molecules in Tissue by MALDI-MS Afforded by Prolonged Sample Storage at Reduced Pressure

Sample preparation is critical for successful MALDI imaging, with sectioning, mounting, matrix application and storage all playing a critical role in the quality of the data obtained. This work demonstrates how an additional factor, prolonged duration of samples in the MALDI chamber may have a significant effect on the imaging of a pharmaceutical (clozapine, [M+H]~(+) (velence) m/z 327) within thin tissue sections. Many factors should be considered when attempting to produce reproducible results via MALDI imaging and apparently identical samples may provide significantly different results if stored for different time periods at reduced pressure.
