Bioconjugation of Lipid Peroxidation Products: A New Role for Vitamin C?

Lipid hydroperoxides are oxidation products of poly-unsaturated fatty acids. Their production is generally thought to be elevated under conditions of oxidative stress. Hydroperoxy octadecadienoic acids (HPODEs), oxidation products of linoleic acid, are subject to degradation into a variety of alpha,beta-unsaturated aldehydes and ketones, many of which are cytotoxic and genotoxic due to their reactivity towards proteins and nucleic acids. We have previously shown that vitamin C mediates the degradation of HPODEs and forms conjugates with reactive aldehydes that result from the degradation of HPODEs (Scheme 1). In this study we examined the fate of 4-hydroxy-2-nonenal (HNE) and 4-oxo-2-nonenal (ONE) in vitamin C-pretreated cells by LC-MS/MS.
