Succinimide, aspartate/iso-aspartate isoforms were characterized from tryptic digest of cetuximab using LC-MS/MS. Three deamidated peptides in the conserved region of heavy and light chains of cetuximab were observed. The extent of deamidation varied depending on the digestion conditions. Cetuximab under recommended storage condition (2-8 deg C) had minimum deamidation, while cetuximab under stress condition (38-42 deg C) had higher deamidation.
