When on-line SPE is fully integrated with LC-MS it can help to optimize the total workflow from raw serum sample to the final data file. For high throughput analysis of relatively small sample batches method development often is experienced to be the time limiting process. For MS automated tuning tools are available to quickly develop a new MS/MS method. Also for LC method development dedicated software is available that can be used to obtain a good separation within a short time frame. For SPE, however, only limited systematic / straightforward procedures can be found. Furthermore, these procedures are considered being tricky and time-consuming. Therefore, a completely new on-line SPE method development approach is explored that is quick and simple
