De novo Sequencing of Tryptic Digests: A Method for Amino Acid Sequencing by Coupling The CAF Method with Affinity-isolation

A method for de novo sequencing of the tryptic digests has been established by effectively using the N-terminal sequencing reagent previously reported (A051428, 53rd ASMS). The digested peptides are sulfonic acid-derivatized with Biotin-SS-reagent, and de novo sequencing of the derivatized peptides are easily performed by chemically assisted fragmentation. HPLC separation of the peptides increases the number of the peptides which are successfully sequenced by MALDI-PSD.
