The development of monoclonal antibodies (Mab) as biotherapeutics is a major focus in the pharmaceutical industry. The nature of the glycosylation of IgG plays a significant role in both its biological function and physiochemical properties. The glycosylation of Mabs can be influenced by cell culture condition, harvesting methods, and the purification process. Regulatory approval recognizes the special properties of such glycosylation, which in turn, requires a particular set of techniques for analysis and characterization. Historically, the structural elucidation of glycans is difficult due to their chemical complexity, different configuration and isoforms. Here we report the use of LC/MSn and OSCAR, an algorithm for assigning oligosaccharide topology from MSn data (Lapadula et al. Anal. Chem. 2005, 77, 6271), for a full and detailed investigation of anti IL-5 carbohydrate structure.
