The need to understand and minimize the side effects of whole brain irradiation (WBI) is exacerbated by the increasing number of patients with brain metastases requiring WBI, -200,000 patients/year,. We hypothesize that WBI leads to activation of the intrinsic renin-angiotensin system (RAS) and induction of proinflammatory mediators regulated via the downstream NADPH oxidase signaling pathway. Irradiating RBMECs increased intracellular ROS generation, NFkB activation, and PAI-1 and ICAM-1 expression, which were ameliorated by inhibition of NADPH oxidase.
